DIRECT ORGANOGENESIS FROM COTYLEDONARY NODES, SHOOT TIPS AND NODAL SEGMENTS OF CUCUMBER (CUCUMIS SATIVUS L.)

AbstractPresent investigation was undertaken for in vitro shoot formation, shoot multiplication, root induction and plantlet establishment on the soil from shoot tips (ST), nodal segments (NS) and cotyledonary node (CN) explants of cucumber (Cucumis sativus L.). The experiments were conducted using Murashing and Skoog’s (1962) medium supplemented with various concentrations and combinations of auxin (NAA, IBA and 2,4-D), cytokinins (BAP, 2-ip) and gibberellic acid (GA3) as growth regulators either alone or in combination with others. A total of 90% contamination free culture was achieved when cucumber explants were treated with 0.05% HgCl2 for 3.5 minutes to kill surface micro organism. Among evaluated direct propagation media, 1 mg/l BAP + 0.1 mg/l GA3 showed highest propagation efficiency from ST and NS, and 1.5 mg/l BAP + 0.2 mg/l GA3 from CN. Micro shoots were rooted well in ½ strength of MS medium supplemented with 0.5 mg/l IBA. Rooted plantlets were successfully established in sandy soil with good survival rate.
