AbstractPlant tissue culture techniques such as multiplication and regeneration are important tools to overcome the challenges of agricultural, medicinal and industrial sectors. Varieties with improved characters were achieved through these processes. To get suitable somaclone of strawberry (Fragaria × ananassa Duch.), leaves of in vitro proliferating shoots were employed on medium containing an auxin and a cytokinin to stimulate callus induction and regeneration in this study. Among the different plant growth regulators (PGR) used for callus induction, the best callus induction media was 3 mg/l NAA + 0.5 mg/l BA. The calli multiplied in NAA+BA supplemented callusing media had a strong regeneration capability. Regeneration media contained
1.5 mg/l BA and 0.75 mg/l NAA demonstrated the best reaction to indirect shoot regeneration. The current study shows that the cytokinin-auxin ratio in callus induction as well as regeneration media are significant determinants in strawberry to develop somatic embryos and subsequently regeneration success. To understand the molecular mechanism of regeneration via somatic embryogenesis, the expression pattern of one of the associated gene i.e. somatic embryogenesis receptor like kinese (SERK) was examined in regenerable and non-regenerable calli of strawberry, and it was observed that regenerable calli (embyogenic) exhibited higher expression of SERK than non regenerable calli.
