IDENTIFICATION OF A BACTERIUM FROM AMERICAN COCKROACH, (PERIPLANETA AMERICANA) BY 16S RRNA GENE SEQUENCING

Abstract

Cockroaches are insects of the order Blattodea. Among them, the best known species is the Periplaneta americana, commonly known as the American cockroaches. They have been found to harbor more than one hundreds of bacteria either on the cuticle or in the gut. We have isolated a bacterium from P. americana that was collected from the student hall of Rajshahi University, Bangladesh. The bacterium was gram negative and antibiotic sensitivity test shows that it is resistant to the antibiotic gentamycin and amoxicillin.We used 16S rRNA gene amplification and sequencing for the identification and taxonomic study of the pathogen. Sequenced PCR product of 16S rRNA gene was 782bp in length. BLAST search of the sequence identified the bacterium as Klebsiella aerogenes strainwith 98% identity. The sequence was deposited in the Gen Bank and accession number of the isolate is KU976965. We have constructed the phylogenetic tree with other strains of K. aerogenes. In the current paper we report a new strain of K. aerogenes from the midgut of P. americana.
