SCREENING OF SYZYGIUM CUMINI FRUIT JUICE FOR ANTI- PROLIFERATIVE ACTIVITY AGAINST EHRLICH ASCITES CARCINOMA (EAC) TUMOR MODEL IN SWISS ALBINO MICE

Abstract

Natural plant derived compounds are always a preferred source of bioactive compounds for developing drugs to cure different diseases specially cancer. Phytochemical constituents present in the Syzygium cumini fruits are well known for its medicinal efficacy although very little is known about its anticancer potential. Therefore, the present investigations were carried out to study the anticancer activity of the S. cumini along with its antioxidant property and cytotoxicity. The antioxidant activity of S. cumini fruit juice was evaluated by DPPH free radical scavenging assay and was compared to BHT standard at different concentrations. S. cumini fruits juice was found to have very high and potent antioxidant property which exhibited a significant dose dependent inhibition of DPPH activity with a IC50value of 44.94 µg/ml. Its cytotoxic activity was evaluated through a widely known biological method called brine shrimp lethality assay where the IC50 value of the juice was 25.38µg/ml. In vivo anticancer activity of S. cumini fruit juice was evaluated against Ehrlich ascites carcinoma (EAC) cells grown in the peritoneum of Swiss albino mice using the dose of 10mg/kg/day. S. cumini fruit juice successfully inhibited the growth of cancer cells by 55.24 % where cell apoptosis induction was confirmed by fluorescence microscopy through DAPI staining. Present investigation suggests that fruit of S. cumini can be a recommended food for cancer patient as well as the purified bioactive compounds present in the juice could be a promising source of anticancer drug lead molecule to cure carcinogenesis.
